Pubertal Recalibration in PI Youth: Associations with Physiology, Mental Health, Behavior, and Inflammation

Abstract

Early life stress (ELS) is associated with a blunted HPA and SNS stress response (McLaughlin et al., 2015), with increased anxiety symptoms (Wiik et al., 2011) and heightened amygdala responses (Tottenham et al., 2011). These findings suggest that the brain’s defense system was calibrated to harsh conditions in early childhood. Previous work demonstrates that puberty opens a window for recalibration of the HPA axis, for better or worse (Gunnar et al., 2019). Is this recalibration window open for the whole defensive system? Study 1 investigates recalibration of the HPA-axis and the SNS in the Bucharest Early Intervention Project (BEIP), and examines early blunted stress reactivity as a transdiagnostic mechanism of later adolescent psychopathology. Study 2 also uses BEIP data and tests whether ELS sensitizes youth to adolescent stressors leading to heightened inflammation as one pathway to poor health. Study 3 uses a different previously-institutionalized (PI) sample to investigate whether increases in cortisol to more normative levels across puberty directly coincide with changes in anxious behaviors and internalizing symptoms. Study 4 follows the same PI sample and examines whether changes in behavior and anxiety occur only during the pubertal transition, or whether they have lasting effects into late adolescence/early adulthood. Moreover, this study tests whether the COVID-19 pandemic, a second significant life stressor, is associated with increased cortisol and greater psychopathology symptoms for PI than comparison youth without ELS histories. This work has important implications for youth experiencing ELS and helps elucidate potential pathways to psychopathology in this population.
