NFκB is constitutively induced in multiple cancer types and is linked to avoidance of apoptosis, increased metastasis and proliferation, and the immune response. NFκB family members are regulated by the Inhibitor of κB (IκB) family members, which hold NFκB inactive in the cytoplasm. Activation of NFκB can be achieved through degradation of IκB by proteasomes, followed by dimerization of NFκB for subsequent nuclear translocation and signaling. PA28γ is a proteasome activator that is frequently dysregulated in cancer and may regulate NFκB activity. PA28γ has been previously associated with NFκB activity by promoting IκB degradation, and also through stabilizing the interaction of NFκB with its transcriptional co-activator p300. Here, we further study potential impacts of PA28γ on the NFκB pathway by assessing the levels of various implicated proteins in PA28γ-/- cells, as well as by assessing differences in NFκB transcriptional activity through downstream gene targets.
